目的:研究褪黑素(MT)对细菌脂多糖(LPS)致宫内感染胎鼠脑组织TNF-α的表达变化,探讨MT对宫内感染胎鼠脑损伤的保护作用。方法:选用妊娠第19天SD大鼠随机分组:对照(S)组、宫内感染(LPS)组和MT处理组,腹腔注射LPS建立大鼠宫内感染模型,LPS组和S组分别腹腔注射LPS 500μg/kg和等容积生理盐水,MT组同时腹腔注射LPS500μg/kg+MT 10 mg/kg。分别于注药后1、6、12、24、48和72 h剖腹取胎鼠,RT-PCR技术检测胎鼠脑组织中TNF-αmR-NA的表达水平。结果:LPS组各小组较S组胎鼠脑组织中TNF-αmRNA含量均明显上升,LPS组在6h达峰值之后缓慢下降;MT组各小组TNF-αmRNA含量均低于LPS组,随时间延长MT组各小组逐渐上升,48 h达峰值,后稍下降。结论:TNF-α表达增加是LPS致宫内感染胎鼠脑损伤的机制之一;MT抑制TNF-α表达,对胎鼠脑损伤有保护作用。
Objective: To study the effect of melatonin (MT) on the expression of tumor necrosis factor-α (TNF-α) in fetal rats induced by intrauterine infection with lipopolysaccharide (LPS), and to explore the protective effect of MT on intrauterine fetal brain injury. Methods: Sprague-Dawley rats were randomly divided into control group (S), intrauterine infection (LPS) group and MT treatment group, intraperitoneal injection of LPS intraperitoneally to establish intrauterine infection model, LPS group and S group were intraperitoneally injected LPS 500μg / kg and an equal volume of normal saline, while MT group was given intraperitoneal injection of LPS 500μg / kg + MT 10 mg / kg. Fetal rats were taken at 1,6,12,24,48 and 72 h after injection, respectively. The expression of TNF-αmR-NA in fetal rat brain tissue was detected by RT-PCR. Results: The content of TNF-αmRNA in brain tissue of LPS group was significantly higher than that of S group. The LPS group decreased slowly at 6h, the content of TNF-αmRNA in each group in MT group was lower than that in LPS group, Each group in the group gradually increased, peaked at 48 h, then decreased slightly. CONCLUSION: The increased expression of TNF-α is one of the mechanisms of LPS-induced intrauterine infection in fetal rat brain. MT inhibits the expression of TNF-α and protects fetal brain from injury.
