目的 研究顺铂对豚鼠耳蜗外毛细胞钙激活的钾电流作用。方法 豚鼠被迅速断头处死 ,暴露耳蜗 ,在解剖显微镜下去除耳蜗侧壁 ,经胰蛋白酶消化并轻轻吹打 ,以获得单离的豚鼠耳蜗外毛细胞。静置后在倒置相差显微镜下观察。应用膜片钳制技术从单离的豚鼠耳蜗外毛细胞获得钙激活的钾通道记录 ,并观察顺铂对它的作用。结果 结果表明该通道活动受到细胞外浓度为 10 -5mol·L-1的顺铂的抑制。结论 顺铂可抑制豚鼠耳蜗外毛细胞的钙激活的钾电流
Objective To study the effect of cisplatin on calcium-activated potassium currents in cochlear outer hair cells of guinea pigs. Methods Guinea pigs were sacrificed promptly by decapitation, the cochlea were exposed, the cochlear wall was removed under a dissecting microscope, trypsinized and gently swirled to obtain isolated guinea pig cochlear outer hair cells. After standing in inverted phase contrast microscope. Calcium-activated potassium channel recording was obtained from isolated guinea pig cochlear outer hair cells using patch-clamp technique and the effect of cisplatin on it was observed. The results showed that the channel activity was inhibited by extracellular concentration of 10 -5 mol·L-1 of cisplatin. Conclusion Cisplatin can inhibit calcium-activated potassium currents in cochlear outer hair cells of guinea pigs
