MK-521是一种血管紧张素转换酶抑制剂,是顺丁烯二酸盐(MK-421)的赖氨酸类似物。和甲巯丙脯酸相比,对健康志愿者和高血压病人更为有效,并有较长的持续作用。本研究比较 MK-521、二氢氯噻嗪和二者合用在轻度和中度原发性高血压病人中的抗高血压疗效,并把血压反应与血管紧张索转换酶抑制引起的生物化学变化相联系。方法:10例轻度和中度原发性高血压且无靶器官损害的病人(治前立位舒张压在95~115毫米汞柱)。研究过程中给150毫当量钠和80毫当量钾的饮食。病人至少停止治疗二周,在给安慰剂治疗1天
MK-521 is an angiotensin-converting enzyme inhibitor and is a lysine analog of maleate (MK-421). Compared with captopril, it is more effective and has a longer lasting effect on healthy volunteers and hypertensive patients. This study compared the antihypertensive effect of MK-521, hydrochlorothiazide, and their combination in patients with mild and moderate essential hypertension and correlates the blood pressure response to the biochemical changes associated with inhibition of angiotensin-converting enzyme . Methods: Ten patients with mild to moderate essential hypertension without target organ damage (antegrade diastolic blood pressure in the range of 95-115 mm Hg). The study gave a diet of 150 milli equivalents of sodium and 80 milli equivalents of potassium. Patients stopped treatment for at least two weeks and placebo for 1 day
