目的:研究栀子配方颗粒对正常昆明小鼠血脂及肝毒性的影响。方法:连续灌胃28d,观察小鼠行为形态学变化;测定小鼠体重与体长,计算Lee’s指数;测定小鼠肝脏湿重并计算肝脏指数;测定血清中TG(甘油三酯)、TC(总胆固醇)、AST(天门冬氨酸氨基转移酶)的含量;对肝脏组织进行病理学检查。结果:栀子配方颗粒浓度1g/kg、3g/kg、6g/kg、9g/kg、12g/kg、16g/kg组的TG均显著降低;血清中TC含量与空白组相比无显著差异;栀子浓度16g/kg组的体长显著增大,Lee’s指数则明显降低;栀子浓度16g/kg组的肝脏指数、AST含量显著升高,与空白组比较P<0.05;光镜下栀子浓度16g/kg组的肝细胞可见明显的肿胀坏死、炎细胞浸润等形态学上的改变。结论:栀子具有显著的降低血清TG作用,但对TC作用不显著;栀子能增加小鼠体长,降低Lee’s指数,16g/kg组作用尤为显著;长期低剂量服用栀子对肝脏无影响,但长期大剂量服用则易造成肝毒性。
Objective: To study the effect of Zhizi formula granule on blood lipid and hepatotoxicity in normal mice. Methods: The morphological changes of mice were observed by gavage for 28 days. The body weight and body length of mice were measured. The Lee’s index was calculated. The liver wet weight was calculated and the liver index was calculated. The levels of TG (triglyceride), TC Total cholesterol), AST (aspartate aminotransferase) content; pathological examination of liver tissue. Results: The TG of 1g / kg, 3g / kg, 6g / kg, 9g / kg, 12g / kg and 16g / kg group of Fructus Gardeniae Granule were significantly decreased. The content of TC in serum was no significant difference compared with the blank group. The body weight of 16g / kg group increased significantly and the Lee’s index decreased significantly. The liver index and AST content of the group with 16g / kg of gardenia increased significantly compared with that of the blank group (P <0.05) The concentration of 16g / kg group of hepatocytes visible swelling and necrosis, inflammatory cell infiltration and other morphological changes. Conclusion: Gardenia jasminoides has a significant effect of lowering serum TG, but its effect on TC is insignificant. Gardenia can increase the body length of mice and decrease Lee’s index, especially in 16g / kg group. Long-term low-dose administration of Gardenia jasminoides has no effect on the liver , But long-term high-dose is easy to cause liver toxicity.
