目的观察缺氧_复氧对大鼠心室肌细胞的损伤及川芎嗪的保护作用。方法在缺氧_复氧条件下 ,测量对照组和不同浓度川芎嗪组对杆形心肌细胞百分比、心肌细胞内K +/Na+浓度比及Ca2 +浓度的影响。结果缺氧_复氧对大鼠心室肌细胞有损伤作用 ,浓度为4μmol·L -1 的川芎嗪对缺氧_复氧损伤细胞有保护作用 ,它能抑制损伤细胞挛缩 ,提高损伤细胞生存率 ,抑制心肌细胞内K +/Na +浓度比的降低、增加细胞内Ca2 +浓度 ,且剂量越大 ,保护作用越好 ,即川芎嗪对缺氧_复氧损伤细胞的保护呈剂量依赖性。结论川芎嗪对缺氧_复氧损伤的大鼠心肌细胞具有保护作用 ,它能显著地对抗缺氧_复氧对心室肌细胞的损伤作用
Objective To observe the injury of rat ventricular myocytes induced by hypoxia and reoxygenation and the protective effect of tetramethylpyrazine. Methods Under hypoxia and reoxygenation conditions, the effects of the control group and different concentration of tetramethylpyrazine group on the percentage of rod-shaped myocytes, the ratio of intracellular K +/Na+ concentration and the concentration of Ca2+ were measured. RESULTS: Hypoxia and reoxygenation caused damage to rat ventricular myocytes. Tetramethylpyrazine at a concentration of 4 μmol·L -1 had protective effects on hypoxia-reoxygenation injured cells. It could inhibit the contracture of damaged cells and increase the survival rate of injured cells. Inhibition of the concentration of K + /Na + in myocardial cells decreased the concentration of intracellular Ca2 +, and the higher the dose, the better the protection, that is, tetramethylpyrazine in a dose-dependent protection of hypoxia and reoxygenation injured cells. Conclusion Tetramethylpyrazine has a protective effect on rat cardiomyocytes injured by hypoxia/reoxygenation. It can significantly inhibit the damage of ventricular myocytes induced by hypoxia and reoxygenation.
