目的探讨肺通口服液治疗鼠肺间质纤维化对弹性蛋白酶(NE)的影响。方法随机将40只SD大鼠分为模型组、治疗组、激素组和对照组各10只。采用BLEMA5建立大鼠肺间质纤维化模型。治疗组给予肺通口服液(250 mg/kg)灌胃,激素组给予氢化考的松(25 mg/kg)肌注,对照组一次性气管内滴入0.9%生理盐水(2 mL/kg),HE、VG染色观察病理学改变,对血清细胞分类计数,用ELISA法测定弹性蛋白酶含量。结果与正常对照组比较,模型组肺泡炎和肺纤维化程度较重,胶原沉积较多,血清细胞总数和细胞学分类比例增多,血清NE含量显著增高(P<0.01);治疗组肺泡炎和纤维化程度较轻,胶原沉积明显减少,细胞总数和细胞学分类比例下降,血清NE含量较模型组和激素组显著降低(P<0.01)。结论肺通口服液明显减少炎性细胞的渗出,降低中性粒细胞弹性蛋白酶含量,从而减轻鼠肺泡炎和肺间质纤维化程度。
Objective To investigate the effect of Feitong Oral Liquid on elastase (NE) in rats with pulmonary interstitial fibrosis. Methods 40 SD rats were randomly divided into model group, treatment group, hormone group and control group. Pulmonary interstitial fibrosis model was established by using BLEMA5. The treatment group was given Tongmai oral liquid (250 mg / kg) intragastrically, while the hormone group was given hydrocortisone (25 mg / kg) intramuscularly. In the control group, 0.9% saline (2 mL / The pathological changes were observed by HE staining and VG staining. The number of serum cells was counted and the content of elastase was determined by ELISA. Results Compared with the normal control group, the alveolitis and pulmonary fibrosis in the model group were more serious, collagen deposition was more, the total number of serum cells and the proportion of cytological classification were increased, and the content of serum NE was significantly increased (P <0.01) Fibrosis was less, collagen deposition was significantly reduced, the total number of cells and cytology classification decreased, serum NE content was significantly lower than the model group and the hormone group (P <0.01). Conclusion Feitong Oral Liquid can significantly reduce the exudation of inflammatory cells and reduce the neutrophil elastase content, thereby reducing the rat alveolitis and interstitial fibrosis.
