目的 :探讨血小板源生长因子 (PDGF)在粉防己碱 (Tet)抑制野百合碱 (MCT)诱导的大鼠肺动脉高压肺血管构形重建中的作用。方法 :建立 (MCT)诱导的大鼠肺动脉高压模型 ,同时以两个实验组加不同剂量的 Tet抑制肺动脉高压 ,用免疫组化方法检测 Tet对 MCT致肺动脉高压大鼠肺小动脉中膜平滑肌细胞 PDGF- A、B表达的影响。结果 :Tet能抑制肺小动脉中膜平滑肌细胞增殖时的 PDGF- B的表达增强 (P <0 .0 1) ,PDGF- A的表达无显著性差异 (P >0 .0 5 )。结论 :Tet抑制 PDGF- B的表达是其抗血管平滑肌细胞增殖、抑制肺动脉高压血管构形重建的分子机制之一。
Objective: To investigate the role of platelet-derived growth factor (PDGF) in the regulation of pulmonary vascular remodeling in rat pulmonary arterial hypertension induced by tetracycline (MCT) induced by tetrandrine (Tet). METHODS: MCT-induced rat pulmonary arterial hypertension model was established. At the same time, two experimental groups were given different doses of Tet to inhibit pulmonary hypertension. Tet was used to detect pulmonary arterial smooth muscle cells in pulmonary artery of MCT-induced pulmonary hypertension rats by immunohistochemistry. Effects of PDGF-A, B expression. RESULTS: Tet inhibited the expression of PDGF-B in the proliferation of pulmonary arterial smooth muscle cells (P < 0.01), and there was no significant difference in the expression of PDGF-A (P > 0.05). Conclusion : Tet inhibits the expression of PDGF-B is one of the molecular mechanisms of its anti-vascular smooth muscle cell proliferation and inhibition of pulmonary artery blood vessel remodeling.
