目的:基于代谢组学技术研究黄芩对正常小鼠血清和肝脏内源性代谢产物的影响。方法:采用基于核磁共振(1H-NMR)代谢组学技术结合多元统计分析及单变量分析手段,比较正常小鼠与黄芩干预小鼠的血清和肝脏内源性代谢物的变化。结果:给予黄芩后小鼠机体内源性代谢物发生了明显变化。与空白组相比,黄芩组小鼠血清中的10种代谢物含量发生变化,如3-羟基丁酸、柠檬酸、胆碱等含量升高,α-葡萄糖和β-葡萄糖含量下降;肝脏中19种代谢物含量发生变化,如谷氨酸、异亮氨酸、亮氨酸、肝糖原、腺苷等含量升高,黄嘌呤等含量降低,这些内源性代谢物的改变涉及机体的糖代谢、脂肪代谢、氨基酸代谢等途径。结论:黄芩对正常小鼠机体代谢的影响不仅与其寒性有关,而且可能与其抗氧化、保肝、降糖等药理作用密切相关,可为黄芩的药性和药理作用机制的深入研究提供参考。
Objective: To study the effect of Scutellaria baicalensis Georgi on endogenous metabolites in serum and liver of normal mice based on metabonomics. Methods: The changes of endogenous metabolites in serum and liver of normal mice and Scutellaria baicalensis Georgi were compared by means of 1H-NMR metabolomics combined with multivariate statistical analysis and univariate analysis. Results: After administration of skullcap, the endogenous metabolites in mice changed significantly. Compared with the blank group, the contents of 10 metabolites in the serum of Scutellariae Radix groups changed, such as 3-hydroxybutyric acid, citric acid, choline and other content increased, α-glucose and β-glucose content decreased; liver 19 kinds of metabolites content changes, such as glutamate, isoleucine, leucine, glycogen, adenosine and other content increased, xanthine content decreased, these changes in endogenous metabolites involved in the body Sugar metabolism, fat metabolism, amino acid metabolism and other ways. Conclusion: The effect of Scutellaria baicalensis Georgi on the metabolism of normal mice is not only related to its coldness, but also may be related to its anti-oxidation, hepatoprotective and hypoglycemic effects, which may provide reference for further study on the pharmacology and pharmacological mechanism of Scutellaria baicalensis.
