目的:建立同时测定云黄连中四种生物碱含量的高效液相色谱方法。方法:采用Agilent ZORBAX Extend-C_(18)(250mm×4.6 mm,5μm),流动相:0.05 mol·L~(-1)KH_2PO_4(含0.2%三乙胺,用H_3PO_4调pH至2.0):乙腈为75:25,流速:1.0 ml·min~(-1),检测波长345 nm,柱温30℃,进样量10μl。结果:药根碱、黄连碱、巴马汀及小檗碱分别在0.012 6~0.126 0、0.025 1~0.251 0、0.012 6~0.126 0及0.137 6~1.376 0μg范围内呈良好线性关系(r≥0.999 4);四种生物碱的平均回收率(n=9)分别为99.49%、99.02%、98.83%、99.08%;RSD分别为1.62%、1.20%、1.36%、1.39%。结论:该方法简便、准确,分离效果好,可用于云黄连的质量控制。
OBJECTIVE: To establish a HPLC method for the simultaneous determination of four alkaloids in Yunhuanglian. METHODS: Agilent ZORBAX Extend-C18 (250 mm × 4.6 mm, 5 μm) was used as the mobile phase. The mobile phase consisted of 0.05 mol·L -1 KH_2PO_4 (containing 0.2% triethylamine and adjusted to pH 2.0 with H_3PO_4) Was 75:25, the flow rate was 1.0 ml · min ~ (-1), the detection wavelength was 345 nm, the column temperature was 30 ℃ and the injection volume was 10μl. Results: There was a good linear relationship between jatrorrhizine, berberine, palmatine and berberine in the range of 0.012 6 ~ 0.126 0,0.025 1 ~ 0.251 0,0.012 6 ~ 0.126 0 and 0.137 6 ~ 1.376 0 μg respectively (r≥ 0.999 4). The average recoveries of four alkaloids (n = 9) were 99.49%, 99.02%, 98.83% and 99.08%, respectively. The RSDs were 1.62%, 1.20%, 1.36% and 1.39%, respectively. Conclusion: The method is simple, accurate, good separation effect, and can be used for the quality control of Coptis chinensis.
