目的建立梅花入骨丹多糖中蛋白含量的测定方法。方法用水提醇沉法提取梅花入骨丹多糖,考马斯亮蓝法测定蛋白含量,比较Sevage法、木瓜蛋白酶-Sevge法、三氯乙酸-Sevage法、三氯乙酸-正丁醇法脱蛋白的效果。结果 Sevage法、木瓜蛋白酶-Sevage法、三氯乙酸-Se-vage法、三氯乙酸-正丁醇法蛋白去除率分别为46.21%、71.43%、81.12%、88.11%,多糖损失率分别为33.73%、18.51%、43.40%、28.15%。结论木瓜蛋白酶-Sevage法脱蛋白较好。
Objective To establish a method for determination of protein content in Plum blossom polysaccharide. Methods Water extraction and alcohol precipitation method was used to extract the polysaccharide of Plum Dulcandra and the content of protein was determined by Coomassie brilliant blue method. The effects of Sevage method, Papain-Sevge method, Trichloroacetic acid-Sevage method and trichloroacetic acid-n-butanol method were compared. Results The protein removal rates of Sevage, Papain-Sevage, Trichloroacetic acid-Se-vage and trichloroacetic acid-n-butanol were 46.21%, 71.43%, 81.12% and 88.11%, respectively. The polysaccharide loss rates were 33.73 %, 18.51%, 43.40%, 28.15%. Conclusion Papain-Sevage deproteinization method is better.
