目的:建立HPLC法测定醋酸奥曲肽中醋酸含量。方法:采用ZORBAX Eclipse XDB-C_(18)色谱柱;流动相:甲醇-磷酸缓冲液(20:1000);检测波长210nm,流速1.5ml·min~(-1),柱温:室温。结果:醋酸浓度在0.05036~1.0072 mg·ml~(-1)范围内线性关系良好(r=0.999 9);平均回收率为100.13%,RSD为0.79%。结论:分析方法可行,准确度高,可用于醋酸奥曲肽中醋酸含量的测定。
Objective: To establish an HPLC method for the determination of acetic acid in octreotide acetate. METHODS: ZORBAX Eclipse XDB-C 18 column was used. The mobile phase was methanol-phosphoric acid buffer (20: 1000). The detection wavelength was 210 nm and the flow rate was 1.5 ml · min -1. Results: The linear range of acetic acid was 0.05036 ~ 1.0072 mg · ml ~ (-1) (r = 0.999 9). The average recovery was 100.13% and the RSD was 0.79%. Conclusion: The method is feasible and accurate. It can be used for the determination of acetic acid in octreotide acetate.
