目的研究依达拉奉对大鼠血肿周围组织丙二醛(MDA)含量、热休克蛋白-70(Hsp-70)及Bcl-2基因表达的影响,探讨依达拉奉的神经保护机制。方法2004年4~12月于大连医科大学附属二院神经内科将健康雄性SD大鼠120只,随机分为假手术组(n=20),ICH治疗组(n=50)和脑出血(ICH)对照组(n=50)。采用Fredrik等建立的自体股动脉取血并向大鼠尾壳核注射方法复制脑出血模型。各组按术后6 h、24 h、48 h、72 h及5 d分为5个亚组,经尾壳核行冠状切片,硫代巴比妥酸比色法测MDA含量,干湿重法测脑含水量,免疫组织化学法测血肿周围组织Bcl-2和Hsp-70表达。结果ICH治疗组各时间点血肿周围脑组织MDA含量及含水量均低于ICH对照组(P<0.05);Bcl-2阳性细胞表达及Hsp-70阳性细胞表达均高于ICH对照组(P<0.01)。结论依达拉奉能降低血肿周围组织MDA含量,上调Bcl-2和Hsp-70表达,增强神经细胞对缺血缺氧的耐受性。
Objective To investigate the effect of edaravone on the malondialdehyde (MDA) content, heat shock protein-70 (Hsp-70) and Bcl-2 gene expression in the perihematoma of rats and the neuroprotective mechanism of edaravone. Methods A total of 120 healthy male SD rats were randomly divided into sham operation group (n = 20), ICH treatment group (n = 50), and intracerebral hemorrhage (ICH) in the Department of Neurology, Second Affiliated Hospital of Dalian Medical University from April 2004 to December 2004. ) Control group (n = 50). The autologous femoral artery established by Fredrik et al was used to take the blood and the model of intracerebral hemorrhage was reproduced by injection into caudate putamen. The rats in each group were divided into 5 subgroups at 6 h, 24 h, 48 h, 72 h and 5 d after operation. The coronal sections were caudate putamen and MDA content was measured by thiobarbituric acid colorimetric method. The wet and dry weight The brain water content was measured by law, and the expression of Bcl-2 and Hsp-70 in tissues around the hematoma was detected by immunohistochemistry. Results Compared with ICH control group, the content of MDA and the content of water in brain tissue around ICH group were significantly decreased at each time point (P <0.05). The expression of Bcl-2 positive cells and Hsp-70 positive cells in ICH group were higher than those in ICH control group (P < 0.01). Conclusion Edaravone can reduce the content of MDA in the perihematomal tissue, up-regulate the expression of Bcl-2 and Hsp-70, and enhance the tolerance of nerve cells to hypoxia and hypoxia.
