目的:优选女贞子的连续超声逆流提取工艺。方法:以齐墩果酸和熊果酸总量为指标,通过正交试验考察超声频率、乙醇体积分数、料液比和温度对提取工艺的影响。采用HPLC测定齐墩果酸和熊果酸含量,流速0.33 mL·min-1,流动相乙腈-甲醇-0.5%冰醋酸溶液(19∶67∶14)。结果:最佳提取工艺为加15倍量80%乙醇于30 kHz提取;齐墩果酸和熊果酸平均总质量分数5.66%,RSD 1.64%。结论:优选的连续超声逆流提取工艺操作简单、科学合理,适合于女贞子工业化大生产。
Objective: To optimize the continuous ultrasonic countercurrent extraction of Ligustrum lucidum. Methods: Taking the total amount of oleanolic acid and ursolic acid as indexes, the effects of ultrasonic frequency, ethanol volume fraction, solid-liquid ratio and temperature on the extraction process were investigated by orthogonal test. The content of oleanolic acid and ursolic acid were determined by HPLC. The flow rate was 0.33 mL · min-1. The mobile phase was acetonitrile-methanol-0.5% glacial acetic acid solution (19:67:14). Results: The optimum extraction process was 15-fold extraction of 80% ethanol at 30 kHz. The average total mass fraction of oleanolic acid and ursolic acid was 5.66% and the RSD was 1.64%. Conclusion: The optimized continuous ultrasonic countercurrent extraction process is simple, scientific and reasonable, suitable for industrial production of Ligustrum lucidum.
