对披针叶黄华(Thermopsis lanceolata)中分离的编号为SP01、RM02的2株蠕形分生孢子内生真菌进行种属分类鉴定。观察真菌的形态,提取真菌DNA,扩增并测定其ITS、GAPDH序列,构建系统发育树,结合形态特征和遗传进化分析结果,并对其进行种属分类。结果表明,SP01菌株的形态特征与凸脐孢属(Exserohilum)中的E.rostratum最为相似,与E.rostratum的遗传进化关系最为密切;RM02菌株的形态特征与弯孢属(Curvularia)中C.spicifera、C.australiensis最为相似,与C.spicifera的遗传进化关系最为密切。确定披针叶黄华中分离的SP01、RM02分别为E.rostratum和C.spicifera。
Two strains of endophytic conidia isolated from Thermopsis lanceolata were isolated and identified as SP01 and RM02. The morphology of the fungi was observed, the fungal DNA was extracted, ITS and GAPDH sequences were amplified and sequenced, and the phylogenetic tree was constructed. The morphological characteristics and genetic evolutionary analysis were combined to classify the fungi. The results showed that the morphological characteristics of SP01 strain were the most similar to E. rostratum in Exserohilum, and the most closely related to the genetic evolution of E. rostratum. The morphological characteristics of the strain RM02 were similar to those in Curvularia. spicifera, C. australiensis most similar, and the genetic evolution of C. spicifera most closely. It was confirmed that SP01 and RM02 isolated from Phlox lappa were respectively E.rostratum and C.spicifera.
