目的观察能量限制(CR)、高能量[包括高糖(HG)、高脂(HP)]培养基对HepG2细胞SIRT7表达的影响,为进一步研究SIRT7的功能及其在相关代谢疾病中的作用机制提供线索。方法采用RT-PCR、Westernblotting等方法检测在不同培养条件下HepG2细胞SIRT7的表达水平。结果与HG[mRNA:(0.3001±0.09661)、蛋白质:(0.2435±0.01240)]培养基相比,CR和HP均上调HepG2细胞SIRT7的mRNA转录水平[(0.4185±0.07617)、(0.4443±0.04717)]和蛋白质表达水平[(0.3300±0.04769)、(0.7834±0.045937)],差异有统计学意义(P<0.05);且CR与HP相比,后者中SIRT7mRNA转录水平和蛋白质表达水平更高,差异有统计学意义(P<0.05)。结论 SIRT7可能参与肝糖和脂代谢调节,影响脂肪肝的形成。
Objective To investigate the effects of energy restriction (CR) and high energy [including high glucose (HG) and high fat (HP)] medium on SIRT7 expression in HepG2 cells. Results Both CR and HP up-regulated mRNA transcription of SIRT7 in HepG2 cells [(0.4185 ± 0.07617), (0.4443 ± 0.04717)] compared with HG [mRNA: (0.3001 ± 0.09661), protein: (0.2435 ± 0.01240) (0.3300 ± 0.04769) and (0.7834 ± 0.045937), respectively, the difference was statistically significant (P <0.05). Compared with HP, the levels of SIRT7 mRNA and protein expression were higher in CR and HP There was statistical significance (P <0.05). Conclusion SIRT7 may be involved in the regulation of hepatic glucose and lipid metabolism, affecting the formation of fatty liver.
