目的:观察局灶性脑缺血损伤大鼠脑组织血管紧张素II(ANG-II)和血清内皮素I(ET-I)水平的变化及补阳还五汤对其的干预作用。方法:以线栓法构建大鼠大脑中动脉栓塞致局灶性缺血损伤模型,实验动物均匀分为正常组、假手术组、模型组、药物(补阳还五汤)组和阳性药(依达拉奉)组。在造模后第7天,ELISA检测损伤侧脑组织ANG-II和血清ET-I水平。结果:与模型组比较,补阳还五汤能显著降低血清ET-I和患侧脑组织ANG-II水平。结论:补阳还五汤对缺血性脑损伤发挥治疗作用可能与其降低血清ET-1和脑组织ANG-II的表达有一定关系。
Objective: To observe the changes of the levels of angiotensin II (ANG-II) and serum endothelin-1 (ET-I) in brain tissue of rats with focal cerebral ischemia and the effect of Buyanghuanwu Decoction on it. Methods: The focal ischemic model of middle cerebral artery occlusion caused by middle cerebral artery occlusion (MCAO) was established by thread occlusion. The experimental animals were evenly divided into normal group, sham operation group, model group, drug (Buyang Huanwu Decoction) group and positive drug Edaravone) group. On the 7th day after the model establishment, the levels of ANG-II and ET-I in the injured side of the brain were measured by ELISA. Results: Compared with the model group, Buyang Huanwu decoction significantly reduced the levels of ET-I and ANG-II in the affected side of the brain. Conclusion: The therapeutic effect of Buyang Huanwu Decoction on ischemic brain injury may be related to the decrease of serum ET-1 and the expression of ANG-II in brain tissue.
