目的 :对真菌 2 2 0 5的代谢产物进行活性跟踪研究 ,分离出具有拮抗白细胞介素 1受体活性的小分子化合物 2 2 0 5A。方法 :采用大孔吸附、溶媒萃取、硅胶柱色谱、ODS反相柱色谱方法进行分离和纯化 ,以酶联免疫法进行活性跟踪 ,通过UV ,IR ,FABMS ,NMR和元素分析进行化合物的结构鉴定。结果 :分离得到的化合物 2 2 0 5A的相对分子质量为 2 80 ,根据波谱解析 ,确定其为已知化合物布雷菲德菌素A。结论 :通过白细胞介素 1受体阻断剂筛选模型筛选出活性物质布雷菲德菌素A ,为首次报道
OBJECTIVE: To investigate the activity of the metabolites of 2,205 fungi, and to isolate small molecule compounds 2 0 5 5A with antagonistic activity on interleukin 1 receptor. METHODS: Macroporous adsorption, solvent extraction, silica gel column chromatography and ODS reverse phase column chromatography were used to separate and purify the compounds. The activity of the compounds was determined by enzyme-linked immunosorbent assay (ELISA) and their structures were identified by UV, IR, FABMS, NMR and elemental analysis . Results: The relative molecular mass of the isolated compound 2020A was 2 80, which was determined to be brefeldin A, a known compound, according to spectroscopic analysis.
