目的观察干细胞白血病基因在再生障碍性贫血和正常骨髓基质细胞中的表达。方法选取再生障碍性贫血标本与正常骨髓基质细胞标本各30份,利用反转录聚合酶链反应酶联免疫吸附方法(RT-PCR-ELISA)对白血病干细胞在悬浮细胞与正常骨髓基质细胞中的表达,并对表达率进行分析。结果正常骨髓基质细胞在骨髓基质干细胞的阳性率高于再生障碍性贫血患者,差异有统计学意义(P<0.05)。干细胞白血病基因在正常骨髓基质细胞的表达水平上更高,明显高于再生障碍性贫血患者,差异具有统计学意义(P<0.05)。结论 SCL基因在再生障碍性贫血患者的骨髓基质细胞中表达较低,这可能同再生障碍性贫血患者造血调控存在异常有关。
Objective To observe the expression of stem cell leukemia gene in aplastic anemia and normal bone marrow stromal cells. Methods A total of 30 samples of aplastic anemia and normal bone marrow stromal cells were selected. The expression of leukemic stem cells in suspension cells and normal bone marrow stromal cells were determined by reverse transcription polymerase chain reaction (RT-PCR-ELISA) Expression, and analysis of the expression rate. Results The positive rate of normal bone marrow stromal cells in bone marrow stromal cells was higher than that in aplastic anemia patients (P <0.05). Stem cell leukemia gene expression in normal bone marrow stromal cells was significantly higher than that of patients with aplastic anemia, the difference was statistically significant (P <0.05). Conclusions The low expression of SCL gene in bone marrow stromal cells of patients with aplastic anemia may be related to abnormalities of hematopoietic regulation in patients with aplastic anemia.
