目的探讨Eph A2和Ephrin A1在宫颈鳞状细胞癌变过程中的表达及其与临床病理因素间的关系。方法应用免疫组织化学方法,检测Eph A2和Ephrin A1在15例宫颈慢性炎症、35例宫颈上皮内瘤变(CIN)和70例宫颈鳞癌组织中的表达,并采用CD 34抗体标记微血管内皮细胞,计算微血管密度(MVD)。分析Eph A 2和Ephrin A1的表达与MVD之间的相关性及其与宫颈鳞癌临床病理特征的关系。结果 70例宫颈鳞癌组织中,Eph A2和Ephrin A1阳性率分别高于CIN组和宫颈慢性炎症组织,差异均有统计学意义(P<0.05)。Eph A2与Ephrin A1的表达与淋巴结转移和侵润深度有关(P<0.05);Eph A2与Ephrin A1的表达呈正相关(r=0.306,P<0.05)。Eph A2和Ephrin A1与年龄、FIGO分期及病理分级之间差异无统计学意义(P>0.05)。宫颈鳞癌的MVD与年龄无关,而与肿瘤分化程度、FI-GO分期、淋巴结转移及局部侵润深度有关(P<0.05)。经Spearman等级相关分析,Eph A2与Ephrin A1的表达与MVD呈显著正相关(P<0.01)。结论 Eph A2和Ephrin A1表达均与宫颈鳞癌的发生、发展密切相关,联合检测有助于宫颈鳞癌早期诊断和预后评价,可以为今后宫颈鳞癌的靶向治疗提供新的靶点。
Objective To investigate the expression of Eph A2 and Ephrin A1 in cervical squamous cell carcinogenesis and its relationship with clinicopathological factors. Methods Immunohistochemistry was used to detect the expression of EphA2 and Ephrin A1 in 15 cases of chronic cervical inflammation, 35 cases of cervical intraepithelial neoplasia (CIN) and 70 cases of cervical squamous cell carcinoma. CD34 antibody was used to label microvascular endothelial cells , Calculate microvessel density (MVD). The relationship between the expression of Eph A 2 and Ephrin A1 and MVD and the clinicopathological characteristics of cervical squamous cell carcinoma were analyzed. Results The positive rates of EphA2 and Ephrin A1 in 70 cases of cervical squamous cell carcinoma were significantly higher than those in CIN group and chronic cervicitis group (P <0.05). The expressions of EphA2 and Ephrin A1 were correlated with lymph node metastasis and invasion depth (P <0.05). There was a positive correlation between EphA2 and Ephrin A1 (r = 0.306, P <0.05). Eph A2 and Ephrin A1 had no significant difference with age, FIGO stage and pathological grade (P> 0.05). The MVD in cervical squamous cell carcinoma was not related to age, but correlated with tumor differentiation, FI-GO stage, lymph node metastasis and depth of local invasion (P <0.05). Spearman rank correlation analysis showed that there was a significant positive correlation between Eph A2 and Ephrin A1 expression and MVD (P <0.01). Conclusion The expressions of Eph A2 and Ephrin A1 are closely related to the occurrence and development of cervical squamous cell carcinoma. Combined detection may be helpful for the early diagnosis and prognosis evaluation of cervical squamous cell carcinoma, which may provide a new target for the future targeted therapy of cervical squamous cell carcinoma.
