目的观察不同线栓头端处理方法及不同插线深度对线栓法制作成年Wistar大鼠大脑中动脉阻断再灌注模型(MCAO/R)造模效果及大鼠存活率的影响。方法依据改良Longa线栓法制作MCAO/R模型,插入不同深度尼龙线制作模型。于24小时、48小时后测定神经功能缺损评分并观察各组大鼠存活率。比较线栓头端蘸蜡处理法和熏香烧灼处理头端法制作MCAO/R模型48小时存活率及神经功能缺损评分。结果第二组、第三组、第四组大鼠24小时和48小时的存活率和神经功能缺损评分与第一组比较,差异有统计学意义(P<0.05)。其中大鼠存活率第一组最高,第四组最低;神经功能缺损评分第一组最低,第四组最高。两种线栓头端处理方法在相同插线深度(1.8 cm、2.0 cm、2.2 cm)的存活率和神经功能缺损评分方面比较,差异无统计学意义(P>0.05)。结论蘸蜡法与熏香烧灼法处理线栓头端对造模成功率无明显影响。线栓插入越深,神经功能缺损评分越高,存活率越低。线栓插入深度2.0 cm更适用于建立大鼠MCAO/R模型。
Objective To observe the effects of different treatment methods and different depths of suture on the model of middle cerebral artery occlusion and reperfusion (MCAO / R) in adult Wistar rats and the survival rate of rat by thread plug method. Methods The MCAO / R model was made according to the modified Longa thread method, and the nylon thread production model with different depths was inserted. After 24 hours and 48 hours, the neurological deficit score was determined and the survival rate of each group was observed. The 48-hour survival rate and neurological deficit score of MCAO / R model were compared with the head-end-dipped wax treatment method and the incense burning cautery head-end method. Results The survival rate and neurological deficit scores of group two, group three and group four at 24 and 48 hours were significantly different from those of group one (P <0.05). Among them, the survival rate of rats in the first group was the highest, the lowest in the fourth group, the lowest in the first group and the highest in the fourth group. There was no significant difference in survival rate and neurological deficit score between the two thread plug head treatments at the same insertion depth (1.8 cm, 2.0 cm, 2.2 cm) (P> 0.05). Conclusions Dipping wax method and incense burning method have no significant effect on the success rate of modeling. The deeper the thread is inserted, the higher the neurological deficit score is, and the lower the survival rate is. Thread insertion depth 2.0 cm is more suitable for establishing rat MCAO / R models.
