目的:研究桑枝中的主要抗氧化活性物质。方法:采用自由基在线液相色谱法和液相色谱-电喷雾质谱联用法,鉴定桑枝中主要抗氧化活性成分,然后采用高效液相色谱-DAD(检测波长320 nm)对其含量进行定量分析。结果:桑枝乙醇提取液中的主要抗氧化活性物质确定为氧化芪三酚。高效液相色谱-DAD定量测定桑枝氧化芪三酚,在14~1 260 mg.L-1具有良好的线性关系(r=1.000),平均加样回收率为98.2%,RSD 1.2%。方法准确灵敏、快速简便。桑枝中富含氧化芪三酚,其含量在品种间存在显著差异。结论:自由基在线液相色谱法结合液相色谱-质谱联用法可定性定量分析桑枝中的抗氧化活性物质。
Objective: To study the main antioxidants in mulberry branches. Methods: The main antioxidative components in mulberry branches were identified by free radical on-line liquid chromatography coupled with liquid chromatography-electrospray ionization mass spectrometry (HPLC-DAD), and the content was quantified by high performance liquid chromatography with DAD (detection wavelength of 320 nm) analysis. Results: The main antioxidant activity in mulberry ethanol extract was identified as oxysterol. High performance liquid chromatography-DAD quantitative determination of mulberry oxygenated stilbenzene, 14 ~ 1 260 mg.L-1 has a good linear relationship (r = 1.000), the average recovery was 98.2%, RSD 1.2%. The method is accurate and sensitive, quick and easy. Mulberry branch is rich in oxygenated triglyceride, the content of significant differences between varieties. Conclusion: Free radical on-line liquid chromatography combined with liquid chromatography-mass spectrometry can be used to qualitatively and quantitatively analyze the antioxidative substances in mulberry branches.
