目的 研究微波辐射与采用水浴加热 ,模拟微波辐射的人视网膜色素上皮细胞在细胞应激和凋亡相关基因转录的差异 ,探讨微波对培养的人视网膜色素上皮细胞基因转录的影响。方法应用基因芯片技术对 2 4 5 0MHz微波辐射、模拟微波辐射的人视网膜色素上皮细胞的mRNA进行检测。结果 在 97条有关应激和凋亡的目的基因中发现有 7条基因转录上调 ,M3116 6基因上调 2 .5 2倍 ,L2 4 12 3基因上调 2 .6 6倍 ,AF0 3970 4基因上调 2 .2 2倍 ,U6 715 6基因上调 2 .0 7倍 ,AF0 4 0 95 8基因上调 2 .13倍 ,NM 0 0 14 2 3基因上调 2 .6 3倍 ,NM 0 0 5 346基因上调 3.6 8倍 ,未检测到转录显著下调的基因。结论 微波辐射可引起培养的人视网膜色素上皮细胞中多个与应激和凋亡有关基因转录上调 ,微波辐射除了辐射引起的热作用外还存在微波辐射本身的作用。
OBJECTIVE: To investigate the differences of transcriptional genes related to stress and apoptosis in human retinal pigment epithelial cells cultured in water bath and simulated microwave radiation, and to investigate the effect of microwave on gene transcription in cultured human retinal pigment epithelial cells. Methods mRNA of human retinal pigment epithelium cells irradiated by microwave radiation and simulated microwave radiation at 2450 MHz was detected by gene chip technology. RESULTS: Seventy-seven genes were found to be upregulated in the 97 genes involved in stress and apoptosis, up-regulated by 2.52 times for the M3116 6 gene, by 2.660-fold by the L2 4 12 3 gene, and by up-regulation of the AF0 39704 gene by 2 .2 2-fold. The U6 7156 gene was up-regulated by 2.07-fold. The AF0 4 0 958 gene was up-regulated by 2.313-fold. The NM 0 0 14 2 3 gene was up-regulated by 2.6-fold and the NM 0 0 5 346 gene was up-regulated by 3.6 8-fold, no significant transcription was detected in the gene. CONCLUSION: Microwave radiation can induce the up-regulation of several transcriptional genes related to stress and apoptosis in cultured human retinal pigment epithelial cells. In addition to the thermal effects caused by radiation, microwave radiation also has the role of microwave radiation itself.
