目的探讨七叶皂苷钠体外抑制Bel-7402细胞增殖中的作用。方法将Bel-7402细胞分对照组和不同浓度七叶皂苷钠组,用MTT法检测细胞增殖,流式细胞术检测细胞周期,膜联蛋白-异硫氰酸荧光素/碘化丙啶双标检测细胞凋亡,Western blot法检测p21和p53蛋白表达。观察各组细胞增殖、细胞凋亡和细胞周期的变化;观察各组p21和p53蛋白表达的变化。结果七叶皂苷钠能抑制Bel-7402细胞的增殖,呈时间和浓度依赖性。当七叶皂苷钠浓度为40μg/ml和60μg/ml时,作用48h后,细胞周期被阻滞在G0/G1期,凋亡率高于对照组(P<0.05);p21和p53蛋白表达水平升高。结论七叶皂苷钠可能通过上调p21和p53蛋白水平诱导Bel-7402细胞凋亡。
Objective To investigate the role of sodium aescinate in inhibiting the proliferation of Bel-7402 cells in vitro. Methods Bel-7402 cells were divided into control group and sodium aescinate group with different concentrations of sodium aescinate, cell proliferation was detected by MTT method, cell cycle was detected by flow cytometry, Annexin-fluorescein isothiocyanate / propidium iodide double-labeled Apoptosis was detected. Western blot was used to detect the expression of p21 and p53 protein. The changes of cell proliferation, apoptosis and cell cycle in each group were observed. The expression of p21 and p53 in each group were observed. Results Sodium aescinate could inhibit the proliferation of Bel-7402 cells in a time and concentration-dependent manner. After treated with sodium aescinate at 40μg / ml and 60μg / ml for 48h, the cell cycle was arrested in G0 / G1 phase and the apoptosis rate was higher than that in the control group (P <0.05). The levels of p21 and p53 protein Rise. Conclusion Sodium aescinate may induce Bel-7402 cell apoptosis by up-regulating p21 and p53 protein levels.
