目的:了解层粘连蛋白γ_2链基因(Lnγ_2)是否在正常大鼠肝组织中表达及其在肝纤维化过程中的作用。方法:以皮下注射四氯化碳(CCl_4)诱发大鼠肝纤维化模型,采用地高辛原位杂交技术对 Lnγ_2 mRNA 在肝组织中表达进行检测,并与Ⅳ型前胶原α_1链(α_1(Ⅳ))基因表达进行比较。结果:正常大鼠肝组织无 Lnγ_2 mRNA 表达,纤维化早期肝组织即有 Lnγ_2mRNA 的异常表达,其表达细胞肝内分布与α_1(Ⅳ)相似。结论:纤维化肝组织的间质细胞能表达 Lnγ_2基因,提示层粘连蛋白异构体基因表达在肝纤维化早期基底膜异常沉积中起一定作用。
Objective: To investigate whether laminin γ 2 chain gene (Lnγ_2) is expressed in normal rat liver tissue and its role in hepatic fibrosis. Methods: The model of hepatic fibrosis was induced by subcutaneous injection of carbon tetrachloride (CCl_4), and the expression of Lnγ_2 mRNA in liver tissue was detected by digoxigenin in situ hybridization. The expressions of α_1 (α_1 Ⅳ)) gene expression were compared. Results: No Lnγ_2 mRNA expression was found in normal rat liver. Lnγ_2 mRNA was abnormally expressed in the early stage of fibrosis, and the intrahepatic distribution was similar to that of α_1 (Ⅳ). CONCLUSION: Lnγ_2 gene can be expressed in interstitial cells of fibrosis liver tissue, suggesting that laminin gene expression may play an important role in the abnormal deposition of early basement membrane in hepatic fibrosis.
