目的观察血液稀释联合局部亚低温对大鼠全脑缺血/再灌注损伤的影响。方法 40只雄性SD大鼠随机分为缺血/再灌注损伤组(I/S)、局部亚低温组(MH)、急性等容性血液稀释组(NH)和局部亚低温联合急性等容性血液稀释组(MHNH),每组10只。血液稀释后3h处死大鼠,采用ELISA法测定血清S-100B的浓度,采用干湿重法测定大脑半球的含水量,应用电镜观察脑皮质神经元超微结构。结果与I/S组比较,其他3组大鼠血清S-100B浓度、脑含水量明显下降(P<0.05),MHNH组最低,皮质神经元损伤最轻。结论急性等容性血液稀释联合局部亚低温后大鼠血清S-100B的浓度明显下降,对缺血/再灌注损伤大鼠有保护作用。
Objective To observe the effects of hemodilution combined with local mild hypothermia on global ischemia / reperfusion injury in rats. Methods Forty male Sprague-Dawley rats were randomly divided into ischemia / reperfusion injury group (I / S), local mild hypothermia group (MH), acute isobaric hemodilution group (NH) and local mild hypothermia Hemodilution group (MHNH), 10 rats in each group. The rats were sacrificed 3h after hemodilution, the concentration of serum S-100B was measured by ELISA, the water content of hemisphere was measured by dry-wet weight method, and the ultrastructure of cerebral cortex neurons was observed by electron microscope. Results Compared with I / S group, the serum S-100B concentration and brain water content in the other three groups were significantly decreased (P <0.05), the lowest in MHNH group, and the lightest in cortical neurons. Conclusions The serum concentration of S-100B in acutely isosceles hemodilution combined with local mild hypothermia is obviously decreased, which has a protective effect on rats with ischemia / reperfusion injury.
