目的研究唐古特红景天舒张大鼠离体肺小动脉的有效部位及机制。方法制备唐古特红景天95%、70%醇提物及水提物,利用HPLC-DAD分析提取物的含量特征。测定不同提取物中总黄酮、总酚酸含量。采用大鼠肺小动脉血管环离体灌流技术,确定药物舒血管作用的有效部位。利用去甲肾上腺素(NE)及氯化钾(KCl)预收缩血管环,初步分析有效部位舒血管作用的机制。结果药物水提物中总黄酮及总酚酸的含量均为最高。水提物中具有末端吸收化合物的种类及含量较醇提物中的少,醇提物中仅化合物的含量差异较大。唐古特红景天舒张肺小动脉的有效部位主要富集于95%醇提物。药物有效部位可以浓度依赖性地舒张肺小动脉(0.76~7.6mg/m L),其舒血管作用与抑制细胞膜受体门控性钙通道的机制有关。结论唐古特红景天95%醇提物是其舒张肺小动脉的主要有效部位。有效部位可能通过抑制细胞膜受体门控性钙通道,减少Ca2+流入血管平滑肌细胞而发挥舒张血管的作用。
Objective To study the effective parts and mechanism of isolated pulmonary arterioles in the Tanggut Rhodiola. Methods 95%, 70% alcoholic extracts and water extracts of Rhodiola sachalinensis were prepared, and the contents of the extracts were analyzed by HPLC-DAD. Determination of different extracts of total flavonoids, total phenolic acid content. Using rat pulmonary arteriolar ring percutaneous perfusion technology to determine the effective vasodilator effect of the site. Norepinephrine (NE) and potassium chloride (KCl) were used to pre-contract the vasculature to initially analyze the mechanism of vasodilatation at effective sites. Results The content of total flavonoids and total phenolic acids in the water extract of the drug were the highest. Water extract with the end of the absorption of compounds of the type and content of alcohol extract less than the alcohol extract of only the content of compounds vary significantly. The effective site of the Tanggut Rhodiola Pulmonary Artery is mainly enriched in 95% ethanol extract. The active site of the drug can relax the pulmonary arterioles (0.76 ~ 7.6mg / m L) in a concentration-dependent manner, and its vasodilatation is related to the mechanism of inhibiting the G-channel of the plasma membrane receptor. Conclusion Tangut Rhodiola 95% alcohol extract is the main effective part of diastolic pulmonary arterioles. The effective site may play a role in relaxing blood vessels by inhibiting the calcium channel of the cell membrane receptor and reducing the influx of Ca2 + into vascular smooth muscle cells.
