人白细胞介素—8cDNA在大肠杆菌表达,制备出重组人白细胞介素—8(rhIL-8),用以治疗大鼠小肠缺血—再灌注(I-R)损伤.发现其明显改善动物低血压状态,抑制血浆内皮素水平的增加.降低小肠髓过氧化物酶活性,显著延长动物存活时间.用rhIL—8灌流离体大鼠肠系膜上动脉,明显抑制缺氧引起的血管内皮素合成与释放.实验结果提示rhIL-8可能通过抑制白细胞粘附和抑制血管内皮素合成与释放.保护组织I—R损伤.
Human interleukin-8 cDNA was expressed in E. coli and recombinant human interleukin-8 (rhIL-8) was prepared for the treatment of intestinal ischemia-reperfusion (IR) injury in rats and was found to significantly reduce the hypotensive state of the animal , Inhibited the increase of plasma endothelin level, decreased the myeloperoxidase activity of the small intestine and prolonged the survival time of the animals significantly.Using the rhIL-8 perfusion of the isolated rat superior mesenteric artery significantly inhibited the synthesis and release of endothelin induced by hypoxia. The experimental results suggest that rhIL-8 may protect the tissue I-R injury by inhibiting leukocyte adhesion and inhibiting the synthesis and release of endothelin.
