目的探讨游离脂肪酸(Free Fatty Acids,FFAs)对人近曲肾小管上皮细胞(HK-2)的影响及作用机制。方法用含有不同浓度(0、125、250、500μmol/L)软脂酸的DMEM-F12培养人近曲肾小管上皮细胞(HK-2),在0h、24h、48h三个时段:MTT法检测不同环境下FFAs对HK-2细胞增殖的影响,流式细胞术检测FFAs对细胞凋亡和细胞周期的影响,免疫细胞化学法检测处理前后HK-2细胞中白细胞介素-β(IL-β)、白细胞介素-8(IL-8)表达的变化。结果 FFAs呈剂量和时间依赖性抑制HK-2细胞的增殖(P<0.05),并可能通过阻滞细胞周期,诱导细胞凋亡(P<0.05)。经FFAs作用后,IL-β、IL-8的表达增加。结论游离脂肪酸有抑制HK-2细胞增殖,促进其凋亡的作用,同时炎性细胞因子IL-β、IL-8表达增多,参与了其发生和发展的过程。
Objective To investigate the effect and mechanism of Free Fatty Acids (FFAs) on human proximal renal tubular epithelial cells (HK-2). Methods Human proximal tubular epithelial cells (HK-2) were cultured in DMEM-F12 containing different concentrations of palmitic acid (0,125,250 and 500μmol / L). At 0h, 24h and 48h, MTT assay The effect of FFAs on the proliferation of HK-2 cells in different environments. The effects of FFAs on apoptosis and cell cycle were detected by flow cytometry. The levels of interleukin-β (IL-β) in HK-2 cells were detected by immunocytochemistry ), Interleukin-8 (IL-8) expression changes. Results FFAs inhibited the proliferation of HK-2 cells in a dose- and time-dependent manner (P <0.05), and blocked the cell cycle and induced apoptosis (P <0.05). After FFAs effect, IL-β, IL-8 expression increased. Conclusions Free fatty acids can inhibit the proliferation and promote the apoptosis of HK-2 cells. Meanwhile, the expression of IL-β and IL-8 is involved in the occurrence and development of HK-2.
