目的利用微卫星分子标记技术对杜仲居群遗传多样性及亲缘关系进行研究。方法对设计的29对EST-SSR引物以及20对已发表的杜仲SSR引物进行筛选,获得的高多态性引物对不同产区杜仲的125个个体进行遗传多样性及亲缘关系分析。结果筛选得到的18对引物共扩增出74条清晰带。Nei’s基因多样性指数H=0.3750,Shannon’s多态性信息指数I=0.5512。基因分化系数Gst=0.1149。湖南石门与桑植之间有一定的亲缘关系,重庆秀山与陕西略阳之间遗传距离最大。结论杜仲居群内具有丰富的遗传多样性,而不同群体间的遗传多样性低,居群间存在较大基因流。
Objective To study the genetic diversity and genetic relationship of Eucommia ulmoides populations using microsatellite markers. Methods 29 pairs of EST-SSR primers and 20 pairs of published Eucommia ulmoides SSR primers were screened. The high polymorphic primers were used to analyze the genetic diversity and genetic relationship among 125 individuals in different regions of Eucommia. Results A total of 74 clear bands were amplified by 18 pairs of primers. Nei’s gene diversity index H = 0.3750, Shannon’s polymorphism information index I = 0.5512. The coefficient of genetic differentiation Gst = 0.1149. Hunan Shimen and Sangzhi have some kinship, Chongqing Xiushan and Shaanxi Lueyang the genetic distance between the largest. Conclusion The Eucommia population is rich in genetic diversity, while the genetic diversity among different populations is low, and there is a large gene flow among populations.
