目的观测2型糖尿病(T2DM)患者尿中足细胞数量的变化,并探讨其相关因素。方法选取124例T2DM患者和25例健康体检者(正常对照组,NC组)。其中T2DM患者分为3组:正常蛋白尿组(NA组),微量白蛋白尿组(MA组),大量蛋白尿组(CP组),应用小鼠抗人肾小球足细胞特异性蛋白Podocalyxin(PCX)单克隆抗体,采用间接免疫荧光技术检测尿中PCX阳性染色细胞,计数尿足细胞数量。ELISA方法检测血清C反应蛋白(CRP)等指标。比较各组的尿中足细胞数量、血CRP水平。结果与糖尿病正常蛋白尿组比较,微量白蛋白尿组、大量蛋白尿组的血CRP、尿足细胞数量均升高(P<0.05)。结论 T2DM患者尿中足细胞数量与尿蛋白相关,并随着血CRP的升高而增加。
Objective To observe the changes of urinary podocytes in type 2 diabetes mellitus (T2DM) and to explore the related factors. Methods 124 cases of T2DM patients and 25 healthy subjects (control group, NC group) were selected. T2DM patients were divided into three groups: normal proteinuria group (NA group), microalbuminuria group (MA group), a large number of proteinuria group (CP group), the application of mouse anti-human glomerular podocyte-specific protein Podocalyxin (PCX) monoclonal antibody. Indirect immunofluorescence assay was used to detect PCX-positive cells in urine and count the number of urinary podocytes. ELISA method to detect serum C-reactive protein (CRP) and other indicators. The number of urinary podocytes and blood CRP levels in each group were compared. Results Compared with diabetic normal proteinuria group, the levels of CRP and urinary podocyte in microalbuminuria group and mass proteinuria group were significantly increased (P <0.05). Conclusion The number of urinary podocytes in T2DM patients is correlated with urinary protein and increases with the increase of blood CRP.
