建立了高效液相色谱测定番石榴中总维生素C含量的方法。样品经过0.1%草酸溶液提取后,用L-半胱氨酸溶液将脱氢抗坏血酸还原成还原型抗坏血酸后测定L-抗坏血酸总量。采用C_(18)柱(4.6 mm×250 mm,5μm),流动相为0.1%草酸溶液,流速1.0 mL/min,检测波长245 nm。L-抗坏血酸在10.0~500μg/mL范围内线性良好,检出限为5.0 mg/kg,回收率为90%~103%,RSD为0.55%。该方法操作简便,灵敏度高,准确性好,适用于番石榴及其他水果中总维生素C的测定。
A method for the determination of total vitamin C content in guava by high performance liquid chromatography was established. After the sample was extracted with 0.1% oxalic acid solution, dehydroascorbic acid was reduced to reduced ascorbic acid by L-cysteine solution and the total amount of L-ascorbic acid was measured. The column was C 4.6 column (4.6 mm × 250 mm, 5 μm). The mobile phase was 0.1% oxalic acid solution and the flow rate was 1.0 mL / min. The detection wavelength was 245 nm. L-ascorbic acid has a good linearity in the range of 10.0-500μg / mL with a detection limit of 5.0 mg / kg, recovery of 90% -103% and RSD of 0.55%. The method is simple and convenient to operate, has high sensitivity, good accuracy and is suitable for the determination of total vitamin C in guava and other fruits.
