目的研究女性盆底功能障碍(PFD)患者重链多肽(MYH)10与肌球蛋白结合蛋白(My BP)-C的表达,探讨PFD的发病机制。方法选取盆底功能正常女性30例(正常组)与PFD患者30例(PFD组),取所有患者耻尾肌标本制作组织匀浆,应用Elisa与实时定量PCR,在蛋白与基因水平对两组患者的MYH10与MyBP-C进行测定。结果 PFD组MYH10表达量为(1.083±0.090),明显低于正常组的(2.798±0.050),差异有统计学意义(P<0.05);MyBP-C表达量为(1.111±0.040),明显低于正常组的(1.794±0.070),差异有统计学意义(P<0.05)。PFD组MYH10的mRNA相对表达量为(2.692±3.334),明显低于正常组的(0±4.111),基因表达减少达6.2倍,差异有统计学意义(P<0.05);MyBP-C的mRNA相对表达量为(1.686±1.848),明显低于正常组的(0±2.528),基因表达减少达3.4倍,差异有统计学意义(P<0.05)。结论 MYH10与My BP-C基因水平的下调可能是女性PFD的发病原因之一。
Objective To investigate the expression of MYH-10 and My BP-C in women with pelvic floor dysfunction (PFD) and to explore the pathogenesis of PFD. Methods Thirty patients with normal pelvic floor function (normal group) and 30 PFD patients (PFD group) were selected. Tissue homogenates were prepared from all the patients with sham tail muscle samples. Elisa and real- The patient’s MYH10 and MyBP-C were measured. Results The expression of MYH10 in PFD group was (1.083 ± 0.090), which was significantly lower than that in normal group (2.798 ± 0.050) (P <0.05). MyBP-C expression was significantly lower In the normal group (1.794 ± 0.070), the difference was statistically significant (P <0.05). The mRNA expression of MYH10 in PFD group was (2.692 ± 3.334), which was significantly lower than that in normal group (0 ± 4.111) and the gene expression was reduced by 6.2 times (P <0.05). The mRNA level of MyBP-C The relative expression level was (1.686 ± 1.848), significantly lower than that of the normal group (0 ± 2.528), and the gene expression was reduced by 3.4 times. The difference was statistically significant (P <0.05). Conclusion The down-regulation of MYH10 and My BP-C gene levels may be one of the causes of PFD in women.
