目的:总结国内外关于肿瘤淋巴管生成的研究进展。方法:应用PubMed和CNKI期刊全文数据库检索系统,以“肿瘤、淋巴管生成和淋巴转移”为关键词,检索2000-01-2010-11的相关文献,共检索到英文文献591篇,中文文献655篇。纳入标准:1)肿瘤淋巴管形态学特点与肿瘤临床病理因素的关系;2)肿瘤淋巴管生成的机制;3)肿瘤淋巴管生成的分子调控机制。根据纳入标准分析31篇文献。结果:肿瘤组织内部淋巴管可能是功能缺陷的,但肿瘤组织内部增多的淋巴管增加了肿瘤淋巴转移的概率,肿瘤组织内部淋巴管密度与肿瘤淋巴结转移相关。肿瘤组织中有功能的淋巴管多数位于肿瘤组织周围,肿瘤组织周围淋巴管密度与肿瘤淋巴结转移相关,肿瘤组织周围淋巴管可能在肿瘤淋巴转移过程中发挥了更为重要的作用。肿瘤淋巴管生成的机制十分复杂,目前尚无定论,有研究认为肿瘤新生淋巴管主要来源于组织中既存的淋巴管内皮细胞。在肿瘤淋巴管生成过程中,许多因子如VEGF-C、VEGF-D、VEGF-A、PDGF、HGF、IGF、FGF-2和Ang等都直接或间接参与其中,但多数研究认为VEGF-C和VEGF-D是肿瘤淋巴管生成中最重要的调控因子。结论:肿瘤淋巴管生成与肿瘤淋巴转移密切相关,抑制肿瘤淋巴管生成能为肿瘤治疗开辟一条新的途径。
Objective: To summarize the research progress on tumor lymphangiogenesis at home and abroad. Methods: Using PubMed and CNKI periodical full-text database retrieval system, with the key words of “tumor, lymphangiogenesis and lymphatic metastasis”, the related literatures of 2000-01-2010-11 were retrieved, 591 English articles were retrieved, 655 articles. Inclusion criteria: 1) the relationship between morphological features of lymphatic vessels and the clinical and pathological factors of the tumor; 2) the mechanism of tumor lymphangiogenesis; 3) molecular regulation of lymphangiogenesis. According to the inclusion criteria, 31 articles were analyzed. Results: The lymphatic vessels in the tumor tissue may be functionally deficient. However, the increased number of lymphatic vessels in the tumor tissue increases the probability of lymphatic metastasis. The lymphatic vessel density in the tumor tissue is correlated with lymph node metastasis. Most of the lymphatic vessels with functions in the tumor tissue are located around the tumor tissue. The lymphatic vessel density around the tumor tissue is related to the metastasis of the tumor. The lymphatic vessels around the tumor tissue may play a more important role in the lymphatic metastasis of the tumor. Tumor lymphangiogenesis mechanism is very complex, is not conclusive, some studies suggest that lymphatic neoplasms derived mainly from tissue existing lymphatic endothelial cells. Many factors such as VEGF-C, VEGF-D, VEGF-A, PDGF, HGF, IGF, FGF-2 and Ang are directly or indirectly involved in lymphangiogenesis. However, most studies suggest that VEGF- VEGF-D is the most important regulatory factor in tumor lymphangiogenesis. Conclusion: Lymphangiogenesis is closely related to tumor lymphatic metastasis. Suppression of tumor lymphangiogenesis can open up a new way for tumor therapy.
