目的探讨胰岛素对血管内皮细胞中miRNA-21的表达和分泌影响。方法用不同浓度胰岛素(0、1、10、50、100和1000nmol/L)干预人脐静脉内皮细胞(HUVECs),48h后收集细胞上清液及细胞,用实时荧光定量PCR方法检测细胞及上清液中miRNA-21的表达情况。结果与0nmol/L胰岛素组相比,10、50、100和1000nmol/L胰岛素组HUVECs中miRNA-21表达均下调(P均<0.05),1000nmol/L胰岛素组细胞中miRNA-21表达最低(P<0.05),但1与0nmol/L胰岛素组HUVECs中miRNA-21表达差异无统计学意义(P>0.05)。与0nmol/L胰岛素组相比,1和10nmol/L胰岛素组HUVECs上清液中miRNA-21表达升高(P<0.05),而50、100和1000nmol/L胰岛素组上清液中miR-NA-21表达降低(P均<0.05),50nmol/L胰岛素组上清液中miRNA-21表达含量最低(P<0.05)。结论不同浓度胰岛素可影响HUVECs中miRNA-21表达,这种变化反映了不同浓度胰岛素对HU-VECs的凋亡和增殖影响。
Objective To investigate the effect of insulin on the expression and secretion of miRNA-21 in vascular endothelial cells. Methods Human umbilical vein endothelial cells (HUVECs) were treated with different concentrations of insulin (0, 1, 10, 50, 100 and 1000 nmol / L) and supernatants and cells were harvested 48 hours later. Serum miRNA-21 expression. Results Compared with 0 nmol / L insulin group, the expression of miRNA-21 in HUVECs of 10, 50, 100 and 1000 nmol / L insulin groups was down-regulated (all P <0.05), and the lowest in the 1000 nmol / L insulin group <0.05). However, there was no significant difference in the expression of miRNA-21 in HUVECs between 1 and 0 nmol / L insulin groups (P> 0.05). Compared with 0 nmol / L insulin group, the expression of miRNA-21 in supernatants of 1 and 10 nmol / L insulin groups increased (P <0.05), while the levels of miR-NA in supernatants of 50, 100 and 1000 nmol / L insulin groups (P <0.05). The expression of miRNA-21 in 50 nmol / L insulin group was the lowest (P <0.05). Conclusion Different concentrations of insulin can affect the expression of miRNA-21 in HUVECs. This change reflects the effect of different concentrations of insulin on the apoptosis and proliferation of HU-VECs.
