目的观察卡托普利对压力超负荷大鼠心肌核因子-κB p65(NF-κB p65)蛋白表达的影响。方法建立腹主动脉缩窄压力超负荷心室肥厚模型,术后4周成功造模并开始给药,疗程为4周。8周后观察各组心脏质量指数和左心室质量指数、苏木素-伊红(HE)染色法观察左心室心肌病理学、Masson染色法观察左心室心肌胶原形态,图像分析测量胶原容积分数(CVF)和血管周围胶原面积(PVCA),免疫组织化学法测定左心室心肌NF-κB p65含量。结果①与假手术组比较,模型组大鼠心脏质量指数和左心室质量指数均明显升高(P<0.01);与模型组比较,卡托普利组则均显著降低(P<0.01)。②与假手术组比较,模型组大鼠CVF和PVCA均明显升高(P<0.01);与模型组比较,卡托普利组则均显著降低(P<0.01)。③与假手术组比较,模型组大鼠心肌NF-κB p65表达明显升高(P<0.01);与模型组比较,卡托普利组则显著降低(P<0.01)。结论卡托普利具有逆转心室肥厚的作用,可能与抑制心肌组织NF-κB p65蛋白表达有关。
Objective To observe the effect of captopril on the expression of NF-κB p65 protein in myocardium of pressure-overload rats. Methods The model of abdominal aortic constriction pressure overload was established. Four weeks after the operation, the model was successfully established and started to be given. The course of treatment was 4 weeks. After 8 weeks, the cardiac mass index and left ventricular mass index of each group were observed, the left ventricular pathology was observed by hematoxylin-eosin staining, the collagen morphology of left ventricle was observed by Masson staining, the collagen content (CVF) was measured by image analysis, And perivascular collagen area (PVCA). The content of NF-κB p65 in left ventricular myocardium was measured by immunohistochemical method. Results ① Compared with the sham operation group, the cardiac mass index and left ventricular mass index of the model group were significantly increased (P <0.01). Compared with the model group, the captopril group was significantly lower (P <0.01). ②Compared with sham operation group, CVF and PVCA in model group were significantly increased (P <0.01); Compared with model group, captopril group was significantly lower (P <0.01). ③Compared with the sham operation group, the expression of NF-κB p65 in the model group was significantly increased (P <0.01). Compared with the model group, the captopril group was significantly decreased (P <0.01). Conclusions Captopril has the effect of reversing ventricular hypertrophy, which may be related to the inhibition of myocardial NF-κB p65 protein expression.
