目的探讨小鼠脂肪源间充质干细胞(ADMSCs)和骨髓源间充质干细胞(BMSCs)表面标记表达的差异。方法分别对小鼠脂肪源和骨髓源间充质干细胞进行成骨、成脂、成心肌分化诱导、鉴定;流式细胞术与细胞免疫荧光法检测CD29、CD44、CD45和CD73的表达并进行比较。结果 BMSCs形态均一,呈长梭形;ADMSCs形态多样,呈梭形、星形、多角形等,胞体较大,表面分泌物较多;两者均可诱导分化为成骨细胞、脂肪细胞和心肌样细胞。流式细胞术检测显示,BMSCs表面分子CD29、CD44和CD73表达率分别为(83.43±1.97)%、(90.33±0.81)%、(2.63±0.42)%;ADMSCs表面分子CD29和CD44的表达率分别为(53.1±1.05)%、(34.8±2.1)%,CD73表达不稳定,在1.8%~19.7%之间波动。两种细胞均不表达造血干细胞标记物CD45。免疫荧光显示,CD73分子与部分CD29、CD44阳性细胞共表达。结论小鼠BMSCs和ADMSCs均具有成脂、成骨、成心肌分化能力,但是在形态上和分子表达上均存在差异。BMSCs的CD44和CD29表达高于ADMSCs;而CD73的表达则相反,CD73在两种细胞的表达均较低,在ADMSCs的表达不稳定。
Objective To investigate the difference of surface markers between mouse adipose derived mesenchymal stem cells (ADMSCs) and bone marrow mesenchymal stem cells (BMSCs). Methods The adipogenic and bone marrow mesenchymal stem cells were induced to osteogenic, adipogenic and cardiomyogenic differentiation respectively. The expressions of CD29, CD44, CD45 and CD73 were detected by flow cytometry and immunofluorescence staining . Results The morphology of BMSCs was long and fusiform. ADMSCs were diverse in shape, such as fusiform, star and polygon, with larger cell body and more surface secretions. Both of them could differentiate into osteoblasts, adipocytes and myocardium Like cells. Flow cytometry showed that the expression rates of CD29, CD44 and CD73 on the surface of BMSCs were (83.43 ± 1.97)% and (90.33 ± 0.81)%, (2.63 ± 0.42)%, respectively (53.1 ± 1.05)%, (34.8 ± 2.1)%, CD73 expression was unstable and fluctuated between 1.8% and 19.7%. Neither cell expressed hematopoietic stem cell marker CD45. Immunofluorescence showed that CD73 molecules were co-expressed with some CD29 and CD44-positive cells. Conclusion Both mouse BMSCs and ADMSCs have adipogenic, osteogenic and myocardial differentiation ability, but there are differences in morphological and molecular expression. The expression of CD44 and CD29 in BMSCs was higher than that in ADMSCs, while the expression of CD73 was opposite. The expression of CD73 in both cells was low and the expression in ADMSCs was not stable.
