目的:研究天麻素对高糖诱导的神经小胶质细胞白介素-1β(IL-1β)、白介素-6(IL-6)表达的影响。方法:将体外培养的小鼠小胶质瘤细胞(BV-2细胞)分成对照组、高糖组、天麻素低、中、高(25,50,100 mg.L-1)剂量组,培养24 h,观察细胞形态。应用酶联免疫吸附法(ELISA)测定培养上清中IL-1,βIL-6蛋白浓度,应用逆转录聚合酶链反应(RT-PCR)方法评价IL-1,βIL-6 mRNA表达。结果:与对照组相比,高糖组培养24 h后,细胞出现聚集,胞体、胞核变大,枝状突起明显,胞浆内颗粒物增多现象,天麻素各剂量组上述改变较轻。与对照组相比,高糖组细胞培养上清液中IL-1,βIL-6浓度,以及细胞内IL-1,βIL-6 mRNA表达明显增加,具有显著统计学差异(P<0.05);而与高糖组相比,天麻素各组细胞培养上清液中IL-1,βIL-6浓度,以及细胞IL-1,βIL-6 mRNA表达均降低,均具有显著统计学差异(P<0.05),其中以中浓度天麻素组最为显著,与高、低浓度组相比,均具有显著统计学差异(P<0.05)。结论:天麻素能抑制高糖诱导的神经小胶质细胞IL-1,βIL-6表达。
Objective: To study the effects of gastrodin on high glucose-induced expression of interleukin-1β (IL-1β) and interleukin-6 (IL-6) in neuronal microglia. METHODS: The in vitro cultured mouse glioma cells (BV-2 cells) were divided into control group, high glucose group, and gastrodin low, medium, and high (25, 50, 100 mg.L-1) dose groups, and cultured for 24 h. Observe the cell morphology. The concentration of IL-1 and β IL-6 protein in the culture supernatant was determined by enzyme-linked immunosorbent assay (ELISA). The expression of IL-1 and β IL-6 mRNA was evaluated by reverse transcription polymerase chain reaction (RT-PCR). RESULTS: Compared with the control group, after 24 hours of culture in the high-glucose group, the cells appeared to aggregate, cell bodies, nucleus became larger, dendrites became more prominent, and the amount of particulate matter in the cytoplasm increased. The above changes in the gastrodin group were lighter. Compared with the control group, the concentrations of IL-1, βIL-6, and the expression of IL-1, βIL-6 mRNA in the supernatant of the high glucose group were significantly increased, with statistically significant differences (P<0.05). Compared with the high glucose group, the concentrations of IL-1, βIL-6, and the expression of IL-1, βIL-6 mRNA in the cell culture supernatants of the gastrodin group were all decreased, with statistically significant differences (P< 0.05), of which the medium-strength gastrodin group was the most significant, compared with the high- and low-concentration groups, there was a significant statistical difference (P <0.05). Conclusion: Gastrodin can inhibit the expression of IL-1, βIL-6 in high glucose induced neuromicroglia.
