目的 探讨比较阿霉素 (ADM)化疗与 43℃加热化疗对人肝癌细胞 - 772 1(以下简称 772 1细胞 )和耐药人肝癌细胞模型 - 772 1/Adm(以下简称 772 1/Adm细胞 )的细胞毒作用 .方法 以体外培养的人肝癌细胞 - 772 1和我们前面培养建立的人肝癌细胞模型 - 772 1/Adm为研究对象 ,采用水浴加温法 ,体外细胞毒试验 (MTT法 ) ,观察阿霉素 (ADM)化疗与加热化疗对细胞的生长抑制规律的影响 ;采用流式细胞技术检测加热对 772 1细胞和 772 1/Adm细胞胞内阿霉素浓度的影响 .结果 1772 1/Adm细胞对阿霉素的敏感性明显低于772 1细胞 ;2加热可以明显提高两种细胞对阿霉素的敏感性 ;3流式细胞检测显示加热可明显提高这两种细胞 ,尤其是772 1/Adm细胞内的阿霉素浓度 .结论 加热可以显著提高人肝癌细胞 - 772 1和耐药人肝癌细胞模型 - 772 1/Adm对阿霉素的敏感性 .
Objective To investigate the comparison between adriamycin (ADM) chemotherapy and 43°C heating chemotherapy for human hepatocellular carcinoma cells - 772 1 (hereinafter referred to as 772 1 cells) and resistant human hepatoma cells model - 772 1/Adm (hereinafter referred to as 772 1/Adm cells) Cytotoxicity. Methods: In vitro culture of human hepatoma cells - 772 1 and the human hepatoma cell model established in our previous study - 772 1/Adm as the subject, using water bath heating method, in vitro cytotoxicity test (MTT method), The effects of adriamycin (ADM) chemotherapy and heated chemotherapy on the growth inhibition of cells were observed. The effect of heating on the intracellular adriamycin concentration of 772 1 cells and 772 1/Adm cells was detected by flow cytometry. Results 1772 1/ The sensitivity of Adm cells to doxorubicin was significantly lower than that of 772 1 cells; 2 heating can significantly improve the sensitivity of both cells to doxorubicin; 3 flow cytometry showed that heating can significantly improve the two cells, especially 772 1/ Admomycin concentration in Adm cells. Conclusion Heating can significantly increase the sensitivity of human hepatoma cells - 772 1 and resistant hepatoma cells model - 772 1/Adm to doxorubicin.
