目的探讨改构型酸性成纤维细胞生长因子(MaFGF)对急性染顺铂(DDP)小鼠肾损害的保护作用及其机制。方法腹腔注射(ip)DDP 10 mg/kg,建立小鼠肾损伤模型,然后每日ipMa FGF 10或20μg/kg,连续7 d。采用全自动分析仪测定肌酐(Scr)、尿素氮(BUN)含量,试剂盒测定丙二醛(MDA)、一氧化氮(NO)含量和超氧化物岐化酶(SOD)、谷胱肽过氧化物酶(GSH-Px)活性,观察肾脏病理学变化。结果 Ma FGF可明显降低血清Scr、BUN及肾皮质MDA、NO含量(P<0.05),引起SOD、GSH-Px活性明显升高(P<0.05),使受损的肾结构明显改善。结论 Ma FGF对顺铂致肾功能损伤有保护作用,其机制可能与抗顺铂的脂质过氧化反应有关。
Objective To investigate the protective effect and mechanism of modified acidic fibroblast growth factor (MaFGF) on renal damage induced by acute cisplatin (DDP) in mice. Methods The model of renal injury in mice was established by intraperitoneal injection of 10 mg / kg DDP and then treated with 10 or 20 μg / kg ipMa FGF daily for 7 days. The content of creatinine (BUN) and creatinine (Scr) and urea nitrogen (BUN) were determined by automatic analyzer. The contents of malondialdehyde (MDA), nitric oxide (NO) and superoxide dismutase (SOD) Oxidase (GSH-Px) activity was observed renal pathological changes. Results Ma FGF could significantly decrease the content of Scr, BUN and the contents of MDA and NO in renal cortex (P <0.05), and increase the activities of SOD and GSH-Px (P <0.05). Conclusion Ma FGF has a protective effect on renal injury caused by cisplatin, and its mechanism may be related to the anti-cisplatin lipid peroxidation.
