目的建立用高效液相色谱法测定盐酸利多卡因胶浆(Ⅰ)含量的方法。方法采用十八烷基硅烷键合硅胶为填充剂,醋酸溶液(取冰醋酸50ml,加水930ml,混匀,用1mol.L-1氢氧化钠溶液调节pH值至3.40)-甲醇(40∶60)为流动相,检测波长为254nm,流速为1.0ml.min-1,柱温为30℃。结果利多卡因在13.6~68.0μg范围内呈现良好的线性关系(r=0.9984),平均回收率为99.9%,RSD为0.7%。结论本方法结果准确,重现性好,可用于盐酸利多卡因胶浆(Ⅰ)的含量测定。
Objective To establish a method for the determination of lidocaine hydrochloride (Ⅰ) by high performance liquid chromatography. Methods The octadecylsilane bonded silica gel was used as filler. Acetic acid solution (50ml of glacial acetic acid and 930ml of water was added, mixed and adjusted to pH = 3.40 with 1mol.L-1 sodium hydroxide solution) -methanol (40:60 ) As the mobile phase, the detection wavelength was 254nm, the flow rate was 1.0ml.min-1, the column temperature was 30 ℃. Results Lidocaine showed a good linearity (r = 0.9984) in the range of 13.6-68.0 μg with an average recovery of 99.9% and a RSD of 0.7%. Conclusion The method is accurate and reproducible. It can be used to determine the content of lidocaine hydrochloride (Ⅰ).
