用基因枪法将雪花莲外源凝集素基因 (gna )转移到优良籼型杂交稻恢复系蜀恢 5 2 7中 ,通过 PCR、PCR- South-ern blotting和 Southern blotting等分子方法检测证明 ,外源基因已稳定遗传到转基因第三代 (T2 )。转基因第一代植株 (T0 )在株高和结实率上与相应的组培、种子实生苗植株相比 ,发生明显的不可遗传的变异。随着繁殖代数的增加 ,转基因植株恢复到与对照植株一致。还讨论了 DNA微量提取法在转基因后代筛选中的运用。
The gna was transferred to the restorer line Shuhui 527 of the indica hybrid rice by gene gun method. The results of PCR, PCR-South-ern blotting and Southern blotting showed that exogenous The gene has been stably inherited into the third generation of transgenic (T2). The first generation of transgenic plants (T0) showed obvious non-heritable variation in plant height and seed setting rate compared with the corresponding tissue culture and seedling plants. As the breeding algebra increased, the transgenic plants returned to the same level as the control plants. Also discussed the use of DNA micro-extraction method in the screening of transgenic progeny.
