目的:检测人白细胞介素-23(IL-23)和白细胞介素-12(IL-12)亚基在各类白血病细胞系中的表达并探讨佛波二酯对IL-23和IL-12亚基表达的调控作用。方法:半定量RT-PCR方法检测13株人白血病细胞系IL-23p19,IL-12p35,IL-12p40 亚基的表达。用佛波二酯处理白血病细胞系,不同时间点取样,半定量RT-PCR方法检测上述亚基的表达变化。结果:检测的大多数白血病细胞系均表达p19 mRNA,p19比p35表达更为普遍;而p40仅在EB病毒感染的LCL和J6-1细胞中测到。佛波二酯可以提高EB病毒阳性细胞系中p19,p35和p40的表达水平。结论:p19可能广泛表达于各类血细胞。佛波二酯可上调EB病毒阳性细胞系p19,p35和p40的表达,表明IL-23与IL-12相类似,其在白血病细胞系中的表达可能与EB病毒感染相关。 OBJECTIVE: To detect the expression of interleukin-23 (IL-23) and interleukin-12 (IL-12) subunits in various leukemia cell lines and to explore the effect of phorbol diester on IL-23 and IL-12 Regulation of subunit expression. Methods: The expression of IL-23p19, IL-12p35 and IL-12p40 subunits in 13 human leukemia cell lines was detected by semi-quantitative RT-PCR. The leukemia cell lines were treated with phorbol diester, samples were taken at different time points, and the expression changes of the above subunits were detected by semi-quantitative RT-PCR. RESULTS: Most of the leukemia cell lines tested expressed p19 mRNA, which was more common than p35, whereas p40 was detected only in Epstein-Barr virus-infected LCL and J6-1 cells. Phorbol diester can increase the expression levels of p19, p35 and p40 in Epstein-Barr virus-positive cell lines. Conclusion: p19 may be widely expressed in various types of blood cells. Phorbol diester upregulated the expression of p19, p35 and p40 in EBV positive cell lines, indicating that IL-23 is similar to IL-12 and its expression in leukemia cell lines may be related to Epstein-Barr virus infection.
