本文主要以有机溶剂法浸提油茶籽外壳粉得到的粗提液作为原料,应用絮凝-沉淀法提纯皂素。在单因素试验的基础上,利用Mintab15.0软件得到回归方程的预测模型并进行响应面分析,确定利用絮凝-沉淀法提纯皂素的最优工艺条件。絮凝试验:加入16%的壳聚糖溶液和皂素损失率为28.46%,杂质损失量为1.39%;沉淀试验:氧化钙加入量1.78%、沉淀温度32.75℃和沉淀时间3.01h,沉淀率为98.99%;释放试验:释放温度52.2℃、释放时间3.17h和碳酸氢铵加入量4.09%,释放率为3.61%。最后,在最优提纯工艺条件下测得纯度为60.4%。
In this paper, organic solvent extraction of Camellia seed oil obtained by the crude extract as raw material, the application of flocculation - precipitation method to purify saponin. On the basis of single factor experiment, the prediction model of regression equation was obtained by Mintab15.0 software and response surface analysis was carried out to determine the optimal process conditions for the purification of saponin by flocculation-precipitation method. Flocculation test: adding 16% chitosan solution and saponin loss rate of 28.46%, impurity loss of 1.39%; precipitation test: the amount of calcium oxide added 1.78%, the precipitation temperature of 32.75 ℃ and the precipitation time of 3.01h, the precipitation rate was 98.99%; release test: release temperature 52.2 ℃, release time 3.17h and ammonium bicarbonate 4.09%, the release rate was 3.61%. Finally, the purity of 60.4% was measured under the optimal purification conditions.
