以全反式维生素甲酸 (RA)处理大肠癌细胞系CCL 187,观察RA对CCL 187细胞生长状态、细胞表面形态、超微结构、分化标志酶碱性磷酸酶活性以及内质网标志酶葡萄糖 6 磷酸酶活性的影响。结果发现 5 μMRA可明显抑制CCL 187细胞的生长 ,扫描电镜显示RA处理后的细胞表面较处理前规则 ,丝状伪足消失 ,出现钝形突起 ,超薄切片经透射电镜观察显示线粒体、内质网等细胞器增多 ,酶活性检测显示内质网标志酶葡萄糖 6 磷酸酶活性增强 ;分化标志酶 -碱性磷酸酶的活性较处理前明显升高 ,第 8天时较对照细胞高 2 3 4%。结果提示RA可诱导大肠癌细胞成熟分化。
To investigate the effects of RA on CCL 187 cell growth status, cell surface morphology, ultrastructure, alkaline phosphatase activity and endoplasmic reticulum marker enzyme glucose 6 (CCL 187) with all-trans retinoic acid (RA) Phosphatase activity. The results showed that 5 μMRA could significantly inhibit the growth of CCL 187 cells. Scanning electron microscopy showed that the surface of RA-treated cells disappeared and the filopodia disappeared and the appearance of blunt protuberances appeared. The ultrastructure of mitochondria and endoplasmic reticulum The number of intracellular organelles increased. The activity of endoplasmic reticulum marker enzyme glucose 6 phosphatase increased. The activity of differentiation marker enzyme - alkaline phosphatase was significantly higher than that before treatment. On the 8th day, the activity was 23.4% higher than that of the control. The results suggest that RA can induce the differentiation of colorectal cancer cells.
