用液体闪烁计数法测定离体再灌注鼠心肌肌质网(SR)和线粒体(Mit)内~(45)Ca~(2+)放射性强度(cpm),比较能量制剂ATP-氯化镁,活性氧自由基清除剂SOD和钙阻滞剂Verapamil对离体缺血(氧)再灌注鼠心肌细胞SR和Mit钙离子浓度的影响。结果表明,ATP-氯化镁,SOD和Verapamil组SR内~(45)Ca~(2+)的cpm均高于对照组cpm(P<0.01或P<0.05),而Mit内均低于对照组cpm(P<0.01)。此3种药物均能增高离体大鼠心肌细胞内SR~(45)Ca~(2+)和降低Mit~(45)Ca~(2+)积聚,从而保护心肌细胞缺血再灌注损伤。
The concentration of ~(45)Ca2+(cpm) in myocardium (SR) and mitochondria (Mit) was measured by liquid scintillation counting in vitro. The energy preparation was compared with ATP-MgCl2, active oxygen free. Effects of Scavengers SOD and Calcium Blocker Verapamil on SR and Mit Calcium Ions Concentration in Isolated Myocardial Cells of Rat Ischemia (Reperfusion). The results showed that the cpm values of ~(45)Ca~(2+) in SR-MgCl2, SOD and Verapamil groups were higher than those in the control group (P<0.01 or P<0.05), while those in the Mit were lower than those of the control group cpm. (P<0.01). All of these three drugs could increase the SR Ca(2+) and decrease the accumulation of mitochondrial Ca(2+) Ca 2+ in isolated rat cardiomyocytes, thus protecting myocardial cells from ischemia-reperfusion injury.
