目的探讨盐酸小檗碱(BER)对胰岛β细胞内质网应激相关凋亡的影响。方法以衣霉素(Tun)诱导胰岛β细胞株MIN6细胞内质网应激,检测BER对细胞增殖、凋亡和胰岛素分泌,及对细胞凋亡相关蛋白ASK1、Caspase-12表达的影响。结果 BER(1、10μmol/L)对内质网应激细胞基础和葡萄糖刺激的胰岛素分泌均无明显影响,可减少细胞凋亡、抑制凋亡相关蛋白ASK1和Caspase-12的表达。结论 BER能减少胰岛β细胞内质网应激相关凋亡,其机制可能与抑制凋亡相关蛋白ASK1和Caspase-12的表达有关。
Objective To investigate the effect of berberine hydrochloride on endoplasmic reticulum stress-related apoptosis in pancreatic β cells. Methods The endoplasmic reticulum stress was induced by tunicamycin (Tun) in MIN6 cells, and the effect of BER on the proliferation, apoptosis and insulin secretion, and the expression of apoptosis-related proteins ASK1 and Caspase-12 were detected. Results BER (1, 10μmol / L) had no effect on the basis of endoplasmic reticulum stress and glucose-stimulated insulin secretion, and decreased the apoptosis and inhibited the expression of apoptosis-related proteins ASK1 and Caspase-12. Conclusion BER can reduce endoplasmic reticulum stress-related apoptosis in pancreatic β-cells, and its mechanism may be related to the inhibition of the expression of apoptosis related proteins ASK1 and Caspase-12.
