目的:研究siRNA对人肺腺癌细胞株A2中Wnt-1基因表达对细胞生物学行为的影响。方法:将siRNA用脂质体转染A2细胞,RT-PCR测定Wnt-1 mRNA表达变化;Western blotting测定Wnt-1蛋白表达的变化;流式细胞仪检测对细胞凋亡的影响。结果:与对照组相比,传染siRNA后,实验组靶基因mRNA表达和蛋白表达均有明显降低,细胞凋亡增加。结论:Wnt-1特异性siRNA能有效干扰A2细胞内其相应基因的表达,而靶基因表达受到抑制后细胞凋亡增加。
Objective: To study the effect of siRNA on the biological behavior of human lung adenocarcinoma cell line A2 in Wnt-1 gene expression. Methods: siRNA was transfected into A2 cells by lipofectamine 2000. The expression of Wnt-1 mRNA was detected by RT-PCR. The expression of Wnt-1 protein was detected by Western blotting. The apoptosis was detected by flow cytometry. Results: Compared with the control group, the mRNA and protein expression of the target gene in the experimental group were significantly decreased and the apoptosis increased after the siRNA was infected. CONCLUSION: Wnt-1 specific siRNA can effectively interfere with the expression of its corresponding gene in A2 cells, while the apoptosis of the target gene is inhibited after its expression is inhibited.
