设定了-10,3,15,25,35℃和自然温度6个水平贮藏东北红豆杉花粉.在贮藏期间采用氯化2,3,5三苯基四唑(TTC)染色法每隔3d测定1次花粉生活力.测定结果表明,东北红豆杉花粉寿命因贮藏温度不同而存在显著差异,温度越高花粉寿命越短.在自然温度下贮藏2周后,50%东北红豆杉花粉便丧失了生活力,7周后全部丧失生活力.而在-10℃冰柜中和3℃冰箱中分别贮藏4个月和2个月后,仍有20%的花粉具有生活力.低温可延长东北红豆杉花粉的贮藏寿命.
Set -10, 3,15,25,35 ℃ and natural temperature of 6 levels of storage of Taxus pollen. Pollen viability was measured every 3 days using 2,3,5-triphenyltetrazolium chloride (TTC) staining during storage. The results showed that the pollen life expectancy of Taxus cuspidata was significantly different due to different storage temperatures. The higher the temperature, the shorter the pollen life span. After 2 weeks of storage at natural temperature, 50% of Taxus cuspidata pollen loses its viability and all lose its viability after 7 weeks. However, 20% of the pollen remained viable after storage for 4 months and 2 months respectively in a -10 ° C freezer and a 3 ° C freezer. Cold can prolong the storage life of Taxus cuspidate pollen.
