目的:观察开心解郁汤对抑郁模型大鼠结肠组织诱导型一氧化氮合酶(iNOS)、谷胱苷肽过氧化物酶(GSH-Px)活性及一氧化氮(NO)含量的影响,探索开心解郁汤保护结肠组织的部分作用机制。方法:将30只SD雄性大鼠随机分为正常组、模型组、开心解郁汤组,每组10只;用慢性应激结合孤养的方式造模21 d。在造模同时,开心解郁汤组给予开心解郁汤ig40 g.kg-1,1次/d,连续21 d。紫外分光光度法检测大鼠结肠组织中iNOS,GSH-Px的活性及NO的含量。结果:与正常组比较,模型组大鼠结肠组织中iNOS活性、NO含量增加,GSH-Px活性明显降低(P<0.05);与模型组比较,开心解郁汤可显著降低抑郁模型大鼠结肠组织中iNOS的活性及NO的含量,提高结肠组织中GSH-Px的活性(P<0.05)。结论:开心解郁汤具有保护抑郁状态下大鼠结肠组织功能的作用,其机制可能与降低结肠组织中iNOS活性及NO含量、提高GSH-Px活性有关。
Objective: To observe the effect of Kaixin Jieyu Decoction on the activity of iNOS and GSH-Px and the content of nitric oxide (NO) in the rat model of depression, Exploring the partial mechanism of action of Happy Relief Decoction in protecting colon tissue. Methods: Thirty male Sprague-Dawley rats were randomly divided into normal group, model group and Kaixin Jieyu Decoction group, with 10 rats in each group. The models were treated with chronic stress and solitary for 21 days. In the modeling at the same time, happy Xieyu soup group given happy Xieyu Tang ig40 g.kg-1, 1 times / d, for 21 days. The activity of iNOS and GSH-Px and the content of NO in rat colon tissue were detected by ultraviolet spectrophotometry. Results: Compared with the normal group, the iNOS activity, the content of NO and the activity of GSH-Px in the model group were significantly decreased (P <0.05). Compared with the model group, Kaixin Jieyu Decoction significantly reduced the number of colon The activity of iNOS and the content of NO in the tissue increased the activity of GSH-Px in colon tissue (P <0.05). CONCLUSION: Kaixin Jieyu decoction can protect the function of colonic tissue in rats with depression. The mechanism may be related to the decrease of iNOS activity, NO content and the activity of GSH-Px in colon.
