以西瓜为材料,采用L27(313)多因素正交设计,研究不同的基因型材料、培养温度、光照强度、培养基中BA、琼脂、蔗糖、IAA和Phl.浓度对试管苗玻璃化的影响.结果表明:培养温度的影响达极显著水平(p<0.01),玻璃化频率15℃>35℃>25℃,15℃与25℃、35℃间差异显著(p<0.05);材料的基因型、光照强度、培养基中BA、琼脂和蔗糖浓度的影响达显著水平,玻璃化频率2n>3n>4n,4n与2n、3n间差异显著;1 000 Lx>2 000 Lx>3 000 Lx,1 000 Lx与2 000、3 000 Lx间差异显著;BA 4 mg/L>2 mg/L>1 mg/L,1 mg/L与2 mg/L、4 mg/L间差异显著;琼脂5 g/L>8 g/L>11 g/L,5 g/L与8 g/L、11 g/L间差异显著;蔗糖10 g/L>30 g/L>50 g/L,10 g/L与30 g/L、50 g/L间差异显著.在继代培养中,降低试管苗玻璃化的综合措施是:培养温度25~35℃,光照强度2 000~3 000 Lx,培养基中BA≤1 mg/L,IAA≥1 mg/L,Phl.≥2 g/L,琼脂8~11 g/L,蔗糖30~50 g/L.
The effects of different genotypes, culture temperature, light intensity, BA, agar, sucrose, IAA and Phl. Concentration on the vitrification of vitro seedlings were studied using L27 (313) orthogonal design with watermelon as material. The results showed that the effects of culture temperature were significant (p <0.01), the vitrification frequency was 15 ℃> 35 ℃> 25 ℃, and the difference was significant between 15 ℃ and 25 ℃, 35 ℃ (p <0.05) Type, light intensity, BA, agar and sucrose concentration in the culture medium reached significant level, the frequency of vitrification was 2n> 3n> 4n, and the difference between 4n and 2n, 3n was significant; 1 000 Lx> 2 000 Lx> There was a significant difference between 1 000 Lx and 2 000, 3 000 Lx; BA 4 mg / L> 2 mg / L> 1 mg / L, 1 mg / L and 2 mg / There was a significant difference between 11 g / L and 5 g / L and 8 g / L, g / L> 8 g / L> 11 g / / L and 30 g / L, 50 g / L significant difference.In the subculture, the comprehensive measures to reduce the vitrification of the tube plantlets are: culture temperature 25 ~ 35 ℃, light intensity 2000 ~ 3000 Lx, medium BA≤1 mg / L, IAA≥1 mg / L, Phl.≥2 g / L, agar 8 ~ 11 g / L and sucrose30 ~ 50 g / L.
